Abstract

The methanol extract of aerial part of Clerodendrum inerme were investigated for anti-inflammatory and analgesic at the dose 200 mg/kg, body weight. The experimental paradigms used were carrageenan, induced pedal edema for anti-inflammatory activity, acetic acid induced writhing methods were used to assess analgesic activity. In acute phase inflammation, a maximum inhibition 60.17% (P < 0.01) was noted at the dose of 200 mg/kg after 3 h of treatment with methanol extract of Clerodendrum inerme (MECI) in carrageenan, induced pedal edema respectively. The extract also produced significant (P < 0.01) analgesic activity in both paradigms. This study exhibits that the methanol extracts of aerial part of Clerodendrum inerme possess anti-inflammatory and analgesic activities.

Abstract

A simple, precise, accurate, rapid and reproducible RP-HPLC procedure was developed for simultaneous estimation of Telmisartan and Pioglitazone in pharmaceutical dosage form at a single wavelength. The mobile phase used was a combination of acetonitrile:0.5% triethylamine (pH 4.5 with orthophosphoric acid) (65:35). The detection of the combined dosage form was carried out at 210 nm and flow rate set 1.3 ml/mins. Linearity obtained in the concentration range of 10 to 50 μg/ml for Telmisartan and 7.5 to 37.5 μg/ml for Pioglitazone with correlation coefficient of 0.9997 and 0.9995 respectively. The result of analysis were validated statistically recovery studies confirmed by the accuracy of the proposed method.

Abstract

A simple, precise, accurate, rapid and reproducible RP-HPLC procedure was developed for simultaneous determination of Cefixime and Ofloxacin in tablet dosage form at a single wavelength. The determination was carried out by using Phenomenox, Gemini C18, Column (50×4.6 mm) with mobile phase used as a combination of acetonitrile:0.05 M Tetra butyl ammonium Hydrogen Sulphate (pH 6.0 with orthophosphoric acid) (35:65). The detection of the combined dosage form was carried out at 290 nm and flow rate set 2.0 ml/min. Linearity obtained in the concentration range of 160 and 240 Cefixime and Ofloxacin with correlation coefficient of 0.9999 and 0.9999 respectively. The result of the analysis were validated statistically and recovery studies confirmed by the accuracy of the proposed method.

Abstract

A sensitive liquid chromatography-electro spray ionization mass spectrometry (LC-ESI-MS) method is developed and validated for rapid determination of Esomeprazole in human plasma. Rabeprazole was used as the internal standard (I.S). Human plasma (80 μl) was first alkalified with 200 ml of sodium bi carbonate (100 mM) and then extracted with 2 ml of Ethyl acetate by vibromax shaker for 15 min. The mixture was centrifuged at 4000 rpm for 15 mins. The supernatant was evaporated to dryness and the residue was reconstituted with water: 0.02% Diethylamine in methanol (3:7 v/v). Samples were separated by using a Gemini C₁₈ reversed phased column (50 mm×4.6 mm I.D, 5μ). Mobile phase consisted of Acetonitrile:water (pH:7.0 with Ammonia) [8:2 v/v]. Esomeprazole and internal standard were measured by electrospray ion source in positive selective ion monitoring mode. The good linearity ranged from 5 ng/ml to 1000 ng/ml and the lowest limit of quantification was 5 ng/ml. The extraction efficiency was approximately 73.4%. The quality control samples were stable when kept at room temperature for 6 hours, -70°C for 29 days and after three freeze- thaw cycles.

Abstract

A simple, precise, accurate and rapid HPTLC method has been developed, and validated for the determination of Pioglitazone HCl and Telmisartan simultaneously, in combined dosage form (15 mg and 20 mg). Pioglitazone HCl and Telmisartan was chromatographed on silica Gel 60F₂₅₄ TLC plate using Toluene:ethyl acetate: methanol (7:2:1 v/v) was used as the mobile phase. The R_f value of Pioglitazone HCl and Telmisartan was 0.56±0.04 and 0.23±0.02 and scanned at 254 nm using camag TLC scanner 3. The applicability of the method for simultaneous determinations of Pioglitazone HCl and Telmisartan was verified by the determination of these compounds in marketed tablets. Results of the analysis were validated statistically, and by recovery studies (99.47-101.36% and 99.40-102.37%). The recovery and RSD values were with in limits given in ICH guidelines, method developments indicates that the suitability of proposed for the routine determination of these compounds in tablets. The validation parameters; linearity the range 30-150 ng/spot and 40-200 ng/spot (r=0.9995 and r=0.9996) sensitivity. The limit of detection was found to be 15 ng/spot and 20 ng/spot. The limit of quantification was found to be 45 ng/spot and 60 ng/spot respectively. The accuracy ((99.88% and 99.33%) and reproducibility were found to satisfactory. The proposed method can be successfully used to determine the drug contents on marketed formulation.
